Оснивање музеја у Сремској Митровици уско је везано за археолошке налазе из времена римског града Сирмијума, кога је Амијан Марцелин (око 330-400.г.н.е.), последњи велики римски историограф, назвао 'многољудном мајком градова' – urbium matris populosae et celebris.

Међутим стицајем друштвених околности у формирању музејске установе одлучујући фактор су били историјски споменици црквених обреда. Првог маја 1946 у „Малој црквици” отворен је Музеј црквене уметности (МЦУ), један од првих те врсте у Демократској Федеративној Југославији, са церемонијалним отварањем 30. јуна исте године.

На основу Упутства Одељења за просвету Главног извршног одбора Аутономне Покрајине Војводине од 2. новембра, којим је регулисана и музеолошка проблематика, 23. новембра 1946. основан је Градски музеј у Сремској Митровици, као музеј комплексног типа са археолошком, етнографском, историјском, уметничком и природњачком збирком. У састав Градског музеја ушла је и поставка МЦУ а на челу истанове је , као добровољни сарадник, био арх.

Бранко Василић, Теренски рад организован је на територији тадашњих пет сремских срезова (сремскомитровачки, шидски, румски, старопазовачки и земунски) преко мреже сарадника – „повереника за старине”. Такав начин рада омогућио је организацију тематских изложби и низ других активности различитог обима и карактера, што је уз остале претпоставке (просторне и кадровске) омогућило да је поставка Градског музеја отворена за јавност 31. маја 1948. године.

Административно-управне промене у Срему, до којих је дошло почетком шездесетих година друге половине XX века, утицале су да је Среско веће Народног одбора сремскомитровачког, 24. децембра 1952. донело одлуку о промени назива Градски музеј у МУЗЕЈ СРЕМА чиме је установа прешла на финансирање од стране Среза. У наредном временском периоду, до краја 2011. било је неколико статусних промена у погледу делатности, надлежности институције и њеног финансирања. Године 1953. у састав Музеја Срема ушла је и новооснована Уметничка галерија, која је уз Сталну музејску поставку, лапидаријум Музеја Срема, Музеј црквене уметности, историјску поставку на простору Спомен парка у Сремској Митровици, сталну изложбу о животу и раду комуниста – робијаша у казнионама Крањевине Југославије Робија – школа револуционара, Меморијалну изложбу «Рохаљ базе» и сталну историјску поставку Срем кроз векове, деценијама презентовала прошлост Срема од најстаријих праисторијских налаза до краја Другог светског рата (1945).

Комплексност музеолошке делатности реализована је уз ангажовање преко 60 музејских радника, у различитим својствима, и бројних музејских сарадника, посебно од почетка првих систематских археолошких ископавања Сирмијума (1957), када су раду археолошких екипа учествовали истакнути југословенски и светски признати археолози чијом је заслугом Музеј Срема постао значајно место на музеолошкој мапи Србије, односно Југославије. Нека од археолошких открића која су у власништву Музеја Срема, као што су: уникатни златници Константина Великог кованих између 317 и 337. године, сунчани сат од мермера с краја I – почетак II века нове ере, златни реликвијар из прве половине V в. н. е. , 85 споменика бенифицијара са Јупитеровог светилишта, златна појасна аварска гарнитура из VII века и низ других значајних музеалија, не само из Археолошког, него и других музејских одељења: Историјског, Одељења историје уметности, Етнолошког одељења и Природњачке збирке чине његову препознатљивост и специфичност на нашим просторима.
Поред презентације музејског материјала на сталним поставкама, значајан вид презентовања свих одељења представљале су и тематске изложбе, од којих су најистакнутије следеће:

Новину у раду Музеја Срема представљало је информативно-пропагaндна активност дефинисана кроз манифестацију «Ноћ музеја», први пут у Сремској Митровици организована 2008. године.
Поред наведених, и других активности реализованих по Закону о заштити културних добара, Музеј Срема је посебну пажњу посветио афирмацији прошлости Срема. Тај вид активности односио се и на организовање различитих предавања и одржавања стручних и научних скупова (уз учешће, како домаћих музејских радника, тако и истакнутих научних радника из земље и иностранства), као што су:

Бранко Василић, син митровачког градоначелника Теодора Василића (1907-1914), основну школу и гимназију завршио је у Сремској Митровици. Студирао је и дипломирао на Техничком факултету у Дрездену (Немачка). Након студија и практичног рада у Дрездену и Паризу, запослио се у државној служби код Грађевинске дирекције у Загребу (1928.г.) где је остао до краја Другог светског рата. Током Другог светског рата илегално је помогао Народноослободилачки покрет. После рата, из породичних разлога, прешао је из Загреба у Сремску Митровицу где се запослио у Окружном народноослободилачком одбору за Срем, при Грађевинском-техничком одсеку. Карактер посла који је обављао, образовни профил и изражен афинитет према културно-историјским споменицима утицали су на његов став да је потребно сакупљати, чувати и презентовати споменичко благо (подаци о инж. арх.

Ирина Непокојчицкај. Значајно место у развоју муеолошке делатности у Сремској Митровици после Другог светског рата припада Ирини Непокојчицкајој. Руска емигранткиња која се као једанаестогодишња девојчица са родитељима настанила у Митровици је после завршене гимназије у Кикинди (1927), безуспешно покушавала да упише сликарску академију у Загребу. Високошколско образовање наставила је у истом граду, на Техничкој високој школи (архитектонски одсек). Након студија (1937), запослила се као цртач у Хидротехничком одељку у Сремској Митровици.
